Laboratory Focus |
| Our laboratory focuses on the role
of growth factors as mediators of the vascular smooth muscle cell proliferation central to
atherogenesis and to restenosis following clinical interventions and uses principles of
tissue engineering to deliver SMC growth inhibitors and/or endothelial cell mitogens to
inhibit myointimal hyperplasia and to induce endothelialization. Our current ongoing
studies include local delivery systems for a battery of recombiant FGF mutants designed
for greater EC specificity, the development of novel bioresorbable polymers to induce
differential growth factor synthesis by phagocytizing macrophages, he delivery of
antisense oligonucleotides and of chimeric mitotoxins to selectively inhibit proliferating
SMCs and the seeding of unmodified and genetically modified ECs onto both
de-endothelialized arteries and onto vascular grafts. Both cell culture and animal models
are used to test efficacy while cell responses are investigated by histologic, biochemical
and molecular analyses.
